There’s nothing quite like planning an experiment, preparing the reagents and looking at the cells under the microscope only to realize that there are a total of four cells in each well.  Luckily, this horror scene is easily avoidable as long as you use this method for calculating how to split cells.

When filming this protocol, we played around with making our own “dolly.”  We set the tripod on a long table and placed magazines under each leg.  Holding the tripod to slide the camera gave us smooth, controlled movement that we ended up using in other videos.  Much cheaper than buying an old dolly from Paramount…
